Response surface methodology was used to optimize the composition of medium for exopolysaccharide production by Enterobacter cloacae VHP-34, a gram-negative coliform bacteria isolated from the costal area of Umargam (20.1738 N-72.7640 E), Gujarat. Using Central Composite Design (CCD) and ANOVA analysis, the optimum values of the tested variables were found to be glucose 10.0%, yeast extract 2.0%, peptone 0.2%, and NaCl 5.0%. This combination yielded exopolysaccharide production of 39.4 g/L, a 1.21-fold increase, by statistical optimization compared with manual method.
